Obecnie skuteczne leczenie jest dost\u0119pne jedynie dla rzadszej, wysi\u0119kowej postaci choroby. Podczas seminarium prof. Christian Grimm przedstawi wyniki bada\u0144 nad rol\u0105 niedotlenienia w rozwoju AMD, wykorzystuj\u0105ce modele mysie, analizy histologiczne, techniki multiomiczne oraz obrazowanie fluorescencyjne z u\u017cyciem biosensor\u00f3w.<\/p>\n<p>Prelegent poka\u017ce, \u017ce przewlek\u0142a aktywacja czynnika HIF1 zaburza metabolizm energetyczny fotoreceptor\u00f3w, prowadz\u0105c do ich obumierania, natomiast nadmierna aktywno\u015b\u0107 HIF2 odpowiada za uszkodzenie kom\u00f3rek RPE. Om\u00f3wi tak\u017ce opracowan\u0105 przez sw\u00f3j zesp\u00f3\u0142 terapi\u0119 genow\u0105 opart\u0105 na wektorach AAV, kt\u00f3ra jednocze\u015bnie hamuje aktywno\u015b\u0107 HIF1 w fotoreceptorach i HIF2 w RPE. W badaniach na zwierz\u0119tach terapia chroni\u0142a struktur\u0119 siatk\u00f3wki i fotoreceptory przez ponad rok, wskazuj\u0105c na nowy, obiecuj\u0105cy kierunek leczenia AMD.<\/p>\n<h3>O prelegencie<\/h3>\n<p>Prof. Christian Grimm jest profesorem okulistyki eksperymentalnej na Uniwersytecie w Zurychu. Doktorat z genetyki molekularnej uzyska\u0142 na Uniwersytecie w Bernie, a nast\u0119pnie prowadzi\u0142 badania nad dojrzewaniem snRNP i transportem ma\u0142ych RNA podczas sta\u017cy naukowych w Szwajcarii i na University of Wisconsin w Madison. W 1997 roku do\u0142\u0105czy\u0142 do Laboratorium Biologii Kom\u00f3rek Siatk\u00f3wki na Uniwersytecie w Zurychu, kt\u00f3rym kieruje od 2006 roku. Dwa lata p\u00f3\u017aniej obj\u0105\u0142 stanowisko profesora okulistyki eksperymentalnej.<\/p>\n<p>Jest autorem ponad 160 publikacji naukowych i sze\u015bciu ksi\u0105\u017cek. Jego badania koncentruj\u0105 si\u0119 na molekularnych mechanizmach obumierania fotoreceptor\u00f3w, sygnalizacji zwi\u0105zanej z niedotlenieniem, metabolizmie siatk\u00f3wki oraz terapii genowej. Kierowa\u0142 licznymi projektami finansowanymi m.in. przez Szwajcarsk\u0105 Narodow\u0105 Fundacj\u0119 Nauki i Uni\u0119 Europejsk\u0105. Za swoje osi\u0105gni\u0119cia otrzyma\u0142 m.in. Nagrod\u0119 Alfreda Vogta, Nagrod\u0119 Pro Retina Germany oraz Pfizer Research Award in Neuroscience.
